چکیده

The synthesis de no2ro of fatty acids by particle-free cell extracts is inhibited by long chain fatty acyl coenzyme A derivatives such as palmityl coenzyme A (l-5). On the other hand, fatty acid synthesis is stimulated by the addition of microsomes (6-9). The enzymes that transfer long chain fatty acyl groups from coenzyme A to glycerol S-phosphate and its derivatives are also located in the microsomes (10-14). These observations suggest that the microsomal stimulation of fatty acid synthesis may be caused at least in part by transfer of acyl groups from long chain fatty acyl coenzyme A to glycerol 3-phosphate. The present report describes the effect of glycerol 3-phosphate on the synthesis of fatty acids. A preliminary account of this work has appeared (15).

Triacylglycerol synthesis in goat mammary gland. The effect of ATP, Mg2+ and glycerol 3-phosphate on the esterification of fatty acids synthesized de novo.

Goat mammary-gland microsomal fraction by itself induces synthesis of medium-chain-length fatty acids by goat mammary fatty acid synthetase and incorporates short- and medium-chain fatty acids into triacylglycerol. Addition of ATP in the absence or presence of Mg2+ totally inhibits triacylglycerol synthesis from short- and medium-chain fatty acids, and severely inhibits synthesis de novo of med...

The control of fatty acid and triglyceride synthesis in rat epididymal adipose tissue. Roles of coenzyme A derivatives, citrate and L-glycerol 3-phosphate.

1. Methods are described for the extraction and assay of acetyl-CoA and of total acid-soluble and total acid-insoluble CoA derivatives in rat epididymal adipose tissue. 2. The concentration ranges of the CoA derivatives in fat pads incubated in vitro under various conditions were: total acid-soluble CoA, 0.20-0.59mm; total acid-insoluble CoA, 0.08-0.23mm; acetyl-CoA, 0.03-0.14mm. 3. An investig...

Metabolic engineering of Mortierella alpina for arachidonic acid production with glycerol as carbon source

BACKGROUND Although some microorganisms can convert glycerol into valuable products such as polyunsaturated fatty acids, the yields are relative low due primarily to an inefficient assimilation of glycerol. Mortierella alpina is an oleaginous fungus which preferentially uses glucose over glycerol as the carbon source for fatty acid synthesis. RESULTS In the present study, we metabolically eng...

Triglyceride synthesis by the small-intestinal epithelium of the pig, sheep and chicken.

1. A comparative study was made of triglyceride synthesis by the intestinal epithelium of pigs, sheep and chickens. In pig and chicken tissue both the glycerol 3-phosphate and the monoglyceride pathway of triglyceride synthesis were operative, but the former pathway predominated in sheep tissue. 2. The fatty acid specificity of the glycerol 3-phosphate pathway was studied in pig and sheep total...

Biosynthesis of Phosphatidic Acid, Lysophosphatidic Acid, Diglyceride, and Triglyceride by Fatty Acyltransferase Pathways in Escherichia coZi*

Fatty acyl coenzyme A-glycerol 3-phosphate, fatty acyl coenzyme A-lysophosphatidic acid, fatty acyl coenzyme A-monoglyceride, and fatty acyl coenzyme A-diglyceride acyltransferase activities were found in a particulate fraction of broken cell preparations of Escherichiu coli. The synthesis of phosphatidic acid and lysophosphatidic acid catalyzed by these enzymes was demonstrated to be dependent...
